We took the train from Florence to Rome and it was only a few hours and very inexpensive (about $30 Canadian, I think). I booked all our tickets online via the Trenitalia web site:
I highly recommend that method of travel as it was safe, quick, and cheap!

Take the train…You’ll kick yourself if you pass up on this opportunity. It also is the best way to get around in any European country but this stretch is rather…good on the eyes.
I took the Eurostar from Florence to Rome. Just go to the main Termini and you can buy tickets from a machine. We were there in early March and it was $66 for two people one way in coach. It wouldn have been $99 each way for us to be in first class. It takes a lot longer to take the local train. Go the day before to assure that you get an assigned seat. I had done that but then decided to change my reservation to something earlier, then my husband and I didn’t get to sit together. I checked out buying tickets online before we left, but it was more expensive than buying them right in the Termini.
when i went to rome from florence, i took a train because it was relatively comfortable and not too expensive.
take the train (www.trenitalia.it), it’s comfortable but not so cheap.
by IC (intercity) it’a about 2h.30′, and 28€ second class and 36€ first class, per person one way
by ES* (Eurostar) it’s 1h40′.. and 33 € second class, 47 € first calss per person, one way.
by car it’s cheaper ( if you are 2 or more you can share oil and motorway costs). i don’t know about car rental costs…
